A CM5 with eMMC doee not have the uSD pins connected to the socket. eMMC and lite modules have different PCB's (as CM4 also had), they are mutally exclusive.
So no, with an eMMC version the uSD interface on the IO-board can't be used. You can add a uSD to GPIO though (you can't boot from that one).
