There's no audible click and the associated LED on the Relay board doesn't light up so the signal can't be getting there.
There is a Register Switch - I've tried multiple positions. When I use the incorrect position then python script returns an error that it can't find the board so I must have it set correctly.

Also, your schematic only shows one Relay lead connected the the realy - the second is directly to Vcc? Is this intentional?
