You will see 5V across the pads without an LED there as there is hardly any current flowing and the voltage drop across the resistor will be close to zero.. When the LED is in place the voltage will be much lower (around 2V, probably) as the resistor will drop around 3V.
I just measured the voltage across the (lit) LED on one of my Zero 2Ws and it was 2V.
